Growth conditions Synchronized, S-phase-enriched cells cells. Cultures in early exponential growth were transferred to medium containing dialyzed serum and 5 μM thymidine. After 2 days of further growth, cells were synchronized by 30 h of isoleucine deprivation, also in medium containing 5 μM thymidine. At 12 h after isoleucine readdition, dNTP pools were extracted from rapidly isolated nuclei.
