Cell line/type as in publication | JB3-B (CHO-S derived mutant for DNA replication) |
Compartment | whole cell |
Experiment Details | |
---|---|
Measurement type | dNTP incorporation assay |
Extraction method | methanol |
Remarks to measurement type | Two-step extraction by North et al modified: 5% trichloroacetic acid was used in place of 5% perchloric acid, acid extracts were neutralized with tri-N-octylamine as in Garret and Santi. |
Growth conditions | Cultured in a 1:1 mixture of DMEM and F12 Ham's medium (DF medium) supplemented with 10% newborn or fetal bovine serum. Cell line is temperature sensitive for DNA replication. Shift from 33 C to 39 C inhibits DNA synthesis. Control is measured from cells kept at 33 C. |
